Japanese researchers report in the online edition of the proceedings of the national academy of sciences that they have discovered a new mechanism for breaking down HA（hyaluronic acid）. The findings will help develop new techniques to fight skin aging and treatments for rheumatoid arthritis and osteoarthrosis.
HA is a colloidal amino polysaccharide acid, which exists in the synovial fluid between tissues, joint head and the vitreous substance inside the eyeball, and plays the role of adhesion, lubrication and protection. HA metabolism is very fast, if the balance between synthesis and decomposition is out of balance, the content of HA in the skin will be reduced, rheumatoid arthritis or osteoarthrosis patients will also appear in the body of HA molecules smaller and reduced content. Using skin fibroblasts and "microarray analysis" and "RNA interference", researchers at keio university and other institutions in Japan found that among tens of thousands of genes, only when the expression of "KIAA1199" gene was inhibited did the HA degradation ability of cells decrease significantly. However, after the "KIAA1199" gene was implanted into cells that could not decompose HA, the cells could regain the ability to decompose HA.
The researchers note that the gene KIAA1199 plays a key role in the breakdown of HA. Abnormal expression of this gene in the synovium of the joint can lead to excessive HA decomposition, which is an important reason for the deterioration of rheumatoid arthritis or osteoarthrosis.
